I need to define an exact point for an isovist view, but do not want the point to affect calculations in Depthmap from the imported dxf file. Is it possible to have lines and points in a dxf file that are used for reference only, and either removed or not used in Depthmap isovist and VGA analysis?

It's possible to have lines in the DXF that are for reference only, if you put them in a different layer which you turn off after importing in depthmapX. However, I'm not sure this helps you as you'll have to move the mouse to turn them off, missing the spot in the process.
I will instead suggest you refer to the the depthmapXcli instructions which will allow you to set a very precise point for whatever isovist you have without resorting to having to find the point with the mouse. You can also do this through something like python/jupyter, here's an example.
