Microscope to WSI registration accuracy study guide line

Glass slides in brown slide case next to microscope.

  1. Group 1: 14,15,16
  2. Group 2: 11,12,13

A.Set up

  1. Launch Matlab 2019a, and visit “C:\000_GITHUB\eeDAP\registrationStudyMicToWSI\src” directory.
  2. Run “Administrator_Input_Screen.m” function

B.Global Registration

  1. Following input file (wsi_slot_x), load glass slides.
  2. Following input file, use correct lens (mag_lres, mag_hres) to do global registration (http://didsr.github.io/eeDAP/000_EEDAP/manualHTML/slide_registration_window.htm?ms=AAA%3D&st=MA%3D%3D&sct=NTAw&mw=MjQw).
  3. Do eyepiece to camera registration.

C. Measure registration accuracy

i. Input user ID "name+time(ddmmyyyy)"

ii. For each ROI

  1. Use joystick move microscope. Make sure there is a small target (eg corner of a cell) in the center of eyepiece (cross of reticle)
  2. Click “Get Global Registration WSI ROI” button in eeDAP
  3. Click “Save Global Registration Result” button in eeDAP
  4. Click “Get Local Registration WSI ROI” button in eeDAP
  5. In imagescope, use “hash annotation mark” to mark the target
  6. Close WSI (not imagescope). Click “yes” to save annotation
  7. Click “Save Local Registration Result” button in eeDAP
  8. Click “Next” button in eeDAP

iii. Finish 10 ROIs measurement for one slide, and then go to the next.
